Kratom Leaf Extract Shows Collagenase Inhibition and Anti-Aging Potential in MDPI Study
Source: Plants (MDPI)
Why it matters
This Chiang Mai University study identifies phenolic compounds in kratom leaves with measurable skin anti-aging properties, expanding the scientific picture of kratom beyond its opioid-active alkaloids. The finding opens a potential research path for kratom as a cosmeceutical ingredient — provided its alkaloid content is removed.
The big picture
Most kratom research focuses exclusively on mitragynine and 7-OH and their opioid receptor activity. This study is among the first to rigorously investigate kratom's flavonoid and phenolic constituents for cosmeceutical uses — a research direction Thailand's 2021 reclassification of kratom as an economic plant actively encourages. The study does not address kratom's safety or legal status for consumption.
Key findings
- Ultrasound-assisted extraction optimized with response surface methodology yielded kratom leaf extract with collagenase inhibition IC50 of 92.63 µg/mL — a measurable anti-aging enzyme activity
- Chlorogenic acid (3.1% w/w) and rutin (1.8% w/w) were quantified as the primary bioactive phenolics driving antioxidant and anti-collagenase activity
- Chlorogenic acid showed the strongest predicted collagenase binding at -12.82 kcal/mol in molecular docking analysis
- Extract maintained human dermal fibroblast cell viability above 80% at all tested concentrations up to 1 mg/mL, indicating low cytotoxicity
- Mitragynine was present at 5.9% w/w of extract — researchers state alkaloid fraction must be removed before any cosmeceutical application
What they say
The researchers concluded that "these findings provide preliminary evidence for the cosmeceutical potential of kratom leaf extract, pending control of its alkaloid content," and noted that chlorogenic acid and rutin together could not fully account for the extract's observed collagenase inhibition, suggesting additional active compounds remain to be identified.
Bottom line
Kratom leaf contains phenolic compounds with genuine anti-aging enzyme activity, but alkaloid removal is a prerequisite before any skin-care application can be developed.
